Abstract
Glukokortikoidi predstavljaju grupu steroidnih hormona koja se upotrebljava u liječenju raznih autoimunih i upalnih bolesti te njihova klinička upotreba proizlazi iz njihove sposobnosti indukcije apoptoze timocitnih i leukemijskih stanica. Iako se glukokortikoidi koriste pri liječenju dugi niz godina, mehanizmi njihovog djelovanja slabo su poznati. Glukokortikoidi mogu djelovati na apoptozu na više načina ali i na više staničnih odjeljaka kao što su stanična jezgra, mitohondriji i stanična ovojnica. Mitohondrijski put aktivacije apoptoze bitan je zbog toga što ovim putem glukokortikoidi mogu regulirati smrt leukemijskih i limfatičkih stanica puno prije nego što to omogućava aktivacija genskom regulacijom u staničnoj jezgri.Ciljevi ovog rada su dokazati translokaciju glukokortikoidnih receptora u mitohondrije, istražiti da li glukokortikoidi imaju utjecaja na regulaciju ekspresije mitohondrijskih gena i mogu li glukokortikoidi aktivirati Bcl-2 obitelj proteina u timocitima.

Glucocorticoids form a group of steroid hormones which are comonly used for treatment of vast majority autoimmune and inflamatory disseases. Their clinical application is a result of their abbility to induce apoptosis in thymocytes and leucemic cells. Although glucocorticoids are already used for many years, exact mechanism oftheir biologycal activity is not well-known. Glucocorticoids can influence apoptosis in different ways and their action is possible in nucleus, mitochodria and cell membrane. Mitochondrial pathway of apoptosis is important because the effect of glucocorticoids can be noticed earlier than their effect on gene regulation in nucleus.Aims of this thesis is to prove translocation of glucocorticoid receptor to the mitochondria, show that glucocorticoids can regulate transcription of mitochondrial genes as well as to confirm activation of Bcl-2 family of proteins in thymocytes by glucocorticoids.
