Abstract
Akumulacija ružmarinske kiseline (RA) u biljkama se često javlja kao dio obrambenog odgovora biljke na stres i napad patogena. Prvi korak biosinteze RA, neoksidativnu deaminaciju L-fenilalanina pri čemu nastaje trans-cimetna kiselina, katalizira enzim fenilalanin-amonij-lijaza (PAL). U staničnoj kulturi ukrasne koprive pojačanoj sintezi RA prethodi pojačana aktivnost PAL. Cilj ovog rada da bio je istražiti utjecaj endogeno sintetiziranog oomicetalnog elicitora ß-kriptogeina na aktivnost enzima PAL i biosintezu ružmarinske kiseline. U tu svrhu uzgojeno je transgeno korijenje ukrasne koprive (Coleus blumei Benth.), u kojem se sintetički gen crypt nalazi pod kontrolom inducibilnog alkoholnog promotora, te tretirano etanolom kako bi se inducirala sinteza -kriptogeina. Ekspresija ß-kriptogeina povećala je aktivnost enzima PAL, ali je uzrokovala značajno smanjenje sadržaja ružmarinske kiseline u linijama transgenog korijenja.

The accumulation of rosmarinic acid (RA) in plants often occurs as part of the plant defense response to stress and pathogen attack. The first step of RA biosynthesis, non-oxidative deamination of L-phenylalanine to trans-cinnamic acid, is catalyzed by the phenylalanine ammonia-lyase (PAL) enzyme. In cell culture of painted nettle enhanced synthesis of RA precedes to increased PAL activity. The aim of this study was to investigate the effect of endogenously synthesized oomycetal elicitor -cryptogein on the PAL enzyme activity and rosmarinic acid biosynthesis. For this purpose transgenic roots of painted nettle (Coleus blumei Benth.) were grown, in which a synthetic gene crypt was under the control of alcohol inducible promoter and treated with ethanol to induce synthesis of -cryptogein. Expression of -cryptogein increased the activity of PAL enzyme but caused a significant decrease of rosmarinic acid content in the genetic lines of transgenic roots.
