Abstract
ß-kriptogein je proteinski elicitor oomicete Phytophthora cryptogea koji pripada skupini ß-elicitina. Elicitori su tvari biološkog ili nebiološkog podrijetla koji, u doticaju sa stanicama viših biljaka, mogu izazvati fiziološke i morfološke promjene, između ostalog induciraju proizvodnju reaktivnih kisikovih tvari te potiču antioksidacijsku obranu biljke. Cilj ovog rada bio je odrediti učinak inducibilne ekspresije gena crypt, koji kodira -kriptogein, na ekspresiju i aktivnost različitih izoenzima superoksid-dismutaze (SOD) u transgenom korijenju ukrasne koprive (Coleus blumei Benth.). U transgenom korijenju gen crypt se nalazi pod kontrolom alkoholnog inducibilnog promotora, tako da je njegova ekspresija inducirana tretmanom s 0,1%-tnim i 1%-tnim etanolom. Korijenje je uzorkovano 3., 7. i 14. dan nakon tretmana. U transgenom korijenju eksprimirane su tri izoenzima SOD-a, dok je izoenzim naveće molekulske mase, SOD 1, eksprimiran samo u korijenju u kome je ekspresija gena crypt inducirana 1%-tnim etanolom. Ekspresija gena crypt uzrokovala je smanjenje aktivnosti izoenzima SOD 3, dok je aktivnost izoenzima SOD 4 bila povećana u odnosu na kontrolu. Izoenzim SOD 4 po zastupljenosti u tkivu odgovara izoenzimu Cu, Zn SOD, koji je inače u biljnom tkivu najzastupljeniji.

ß-cryptogein is the protein elicitor of Oomycetes Phytophthora cryptogea who belongs to the group of ß-elicitors. Elicitors can be of biological or non-biological origin and, in contact with the cells of higher plants, can cause physiological and morphological changes. Among other things, they can induce the production of reactive oxygen species and stimulate antioxidant defense of plants. The purpose of this study was to determine the effect of inducible gene expression crypt, which is encoded by ß-cryptogein, on the expression and activity of different isoenzymes of superoxide dismutase (SOD) in transgenic roots decorative nettle (Coleus blumei Benth.). In the transgenic roots gene crypt is under the control of an inducible promoter of alcohol, so that its expression is induced by treatment with 0.1 and 1% ethanol. The roots were sampled 3, 7 and 14 days after the treatment. In the transgenic roots expressed three SOD isoenzymes, while the isoenzyme with the biggest molecular weight SOD 1 is expressed only in the roots in which the gene expression was induced by 1% ethanol. Crypt gene expression caused a decrease in the activity of SOD isoenzymes 3, while the activity of SOD isoenzymes 4 was increased compared to the control. According to its concentration in the tissue SOD isoenzyme 4 responds to the isoenzyme Cu, Zn SOD, which is normally the most common isoenzyme of SOD in a plant tissue.
