Abstract
Gvajakol-peroksidaze (GPOD) postoje u brojnim izooblicima, koji igraju važnu ulogu u oksidacijskom stresu kod biljaka. ß-kriptogein, protein kojeg izlučuje fitopatogena oomiceta Phytophthora cryptogea između ostalog u biljaka uzrokuje stvaranje reaktivnih kisikovih jedinki, te na taj način potiče antioksidacijsku obranu biljke. Cilj ovog rada bio je istražiti utjecaj elicitora ß-kriptogeina na ekspresiju i aktivnost izoenzima GPOD-a u transgenom korijenju ukrasne koprive (Coleus blumei Benth). U tu svrhu uzgojena je O3 linija transgenog korijenja koja sadrži gen crypt pod kontrolom alkoholnog inducibilnog promotora, tako da je njegova ekspresija inducirana tretmanom s 0,1 i 1%-tnim etanolom. Korijenje je uzorkovano 3., 7. i 14. dan nakon tretmana. U transgenom korijenju ukrasne koprive eksprimiran je jedan izooblik GPOD-a. Inducibilna ekpresija gena crypt u transgenom korijenju uzrokuje povećanu aktivnost izoenzima GPOD, jednog od značajnih antioksidacijskih enzima, što upućuje na značajnu ulogu endogeno sintetiziranog ß-kriptogeina u povećanju antioksidacijskog statusa transgenog korijenja ukrasne koprive.

Guaiacol peroxidases (GPOX) exist in a number of isoforms, which play an important role in the oxidative stress in plants. ß -cryptogein, a protein secreted by phytopathogenic oomycete Phytophthora cryptogea inter alia causes the formation of reactive oxygen species in plants, and thus stimulates the antioxidative defense of the plants. The aim of this study was to investigate the effect of elicitor ß-cryptogein on the expression and activity of GPOX isoenzymes in transgenic roots of painted nettle (Coleus blumei Benth). For this purpose the O3 line of transgenic roots was cultivated containing a gene crypt under the control of alcohol inducible promoter so that its expression is induced by treatment with 0.1 and 1% ethanol. The roots were sampled 3, 7 and 14 days after the treatment. In the transgenic roots of painted nettle one GPOX isoform was expressed. Inducible crypt gene expression in the transgenic roots caused increased activity of GPOD isoenzymes, one of the major antioxidant enzymes, suggesting an important role of endogenously synthesized -cryptogein in the increase of antioxidant status in transgenic roots of painted nettle
