Abstract
Huntingtonova bolest jedna je od poznatijih nasljednih bolesti živčanog sustava. Karakteriziraju je simptomi kao što su nekontrolirani motorni pokreti te postepeno kognitivno propadanje koji uglavnom nastupaju u srednjoj životnoj dobi. Mutacija DNA uzrokovana višestrukim ponavljanjem tripleta dušičnih baza CAG uzrok je oboljenja. Dijagnoza bolesti postavlja se genetičkim testiranjem, a liječenje se zasad temelji na ublažavanju simptoma. Iako su biokemijski putevi bolesti višestruko istraženi, stručnjaci još uvijek dolaze do novih spoznaja o pojedinim regulatornim molekulama. IкB kompleks je složena proteinska struktura, a po funkciji enzim građen od tri podjedinice. U organizmu je sveprisutan no u neuronima CNS kod HB pacijenata ključan je u signalnim putevima bolesti. IкBβ podjedinica kompleksa važna je u procesu imunološkog odgovora, a kod Huntingtonove bolesti inhibicija ili stimulacija aktivnosti IкBβ direktno utječe na razvoj simptoma pa i napredak same bolesti.
Abstract (english)
Huntington's disease represents a well known hereditary disorder of nervous system. It is characterized by symptoms such as uncontrolled motor movements and gradual cognitive decay that generally occur in middle age. Cause of the disease is mutation that happens as a result of multiple repeated CAG nitrogen bases. Diagnosis is set by genetic testing, and treatment is based on the relief of the symptoms. Although the biochemical pathways of the disease have been researched, experts are still coming up with new findings about some regulatory molecules. The IкB complex is a compounded protein structure of three subunits, with the function of an enzyme. In organism is ubiquitous, but in CNS neurons of HB patients is crucial in signal pathways of the disease. The IкBβ subunit of complex is important in the immune response processes and at Huntington's disease, inhibition or stimulation of IкBβ activity directly affects the development of symptoms and progression of the illness.
