Abstract
Pročišćeni imunoglobulin G iz seruma brojnih životinjskih vrsta ima široku primjenu u medicinskim i biološkim istraživanjima, kliničkoj dijagnostici i industriji. Cilj rada je odrediti optimalnu metodu za određivanje ukupne koncentracije proteina u serumu, te izolirati, pročistiti i identificirati IgG iz seruma svinja. Za određivanje ukupne količine proteina u serumu korištene su tri različite spektrofotometrijske metode: Lowry, Bradford i UV 280 nm metoda. UV 280 nm metoda odabrana je za daljnje etape istraživanja zbog osjetljivosti i jednostavnosti provedbe. Izolacija i pročišćavanje IgG-a iz seruma postignuto je isoljavanjem pomoću amonijevog sulfata u rasponu zasićenja 10% - 100% i gel filtracijom. Elektroforetskom i Western blot analizom identificirani su nativni IgG te IgG teški i IgG laki lanac, te je procijenjeno da je najveća koncentracija proteina u frakciji dobivenoj isoljavanjem 30%-tnom otopinom amonijevog sulfata. Zaključeno je da se za izolaciju IgG-a iz seruma treba koristiti raspon zasićenja 30% - 50%.

Purified immunoglobulin G from sera of many animal species is widely used in medical and biological research, clinical diagnosis and industries. The aim of study is to determine optimal method for total protein estimation and to isolate, purify and identify IgG from porcine serum. Three different spectrophotometric methods have been used for total protein estimation: Lowry, Bradford and UV 280 nm method. UV 280 nm method was chosen for further steps of research because of its simplicity and sensitivity. Isolation and purification of IgG was accomplished by ammonium sulfate precipitation in saturation range 10% - 100% and gel filtration. Native IgG and IgG heavy and IgG light chain were detected and identified by native and SDS electrophoresis and Western blot. Further, it was esimated that concentration of IgG is higest in fraction obtained with 30% saturated ammonium sulfate. It is concluded that optimal saruration range for IgG isolation is 30% - 50% saturated ammonium sulfate.
