Abstract
Ptijalin je enzim koji se nalazi u sastavu sline gdje katalizira hidrolizu škroba. Nehidrolizirani škrob, čija je koncentracija obrnuto proporcionalna koncentraciji ptijalina, stvara kompleks s trijodidom smanjujući pritom početnu koncentraciju trijodida. Promjena koncentracije trijodidnog iona izaziva promjenu vrijednosti omjera koncentracija redoks para trijodid/jodid, što prema Nernstovom izrazu uzrokuje promjenu redoks potencijala redoks senzora. Ta je promjena u direktnoj korelaciji s koncentracijom ptijalina. U radu je uporabom platinskog redoks senzora provedena direktna potenciometrijska metoda za određivanje aktivnosti ptijalina u biološkim uzorcima sline. U sklopu rada ispitan je utjecaj stresora na aktivnost ptijalina u slini.

Salivary alpha amylase (sAA9, an enzyme present in human saliva, catalyses the breakdown of starch. The nonhydrolysed starch, which amount is inversely proportional to the sAA concentration, forms a complex with triiodide reducing accordingly initial triiodide concentration. The change of triiodide ion concentration causes the change of triiodide/iodide redox-couple ratio, which results with the changes of the changes of the electrode potential of a redox-sensor. The changes are directly correlated with sAA concentration. In this paper direct potentiometric method for determination of sAA activity in human saliva was performed. Additionally, sAA response to the different stressors have been investigated.
