Abstract
Pesticidi su toksični spojevi koji imaju štetne učinke na ljudsko zdravlje i okoliš. U današnje vrijeme, organofosfatni spojevi spadaju u najkorišteniju vrstu pesticida. Veliki udio u njihovoj ukupnoj primjeni ima klorpirifos (CPF), organofosfatni pesticid širokog spektra. Glavni mehanizam toksičnosti organofosfatnih spojeva je ireverzibilna inhibicija acetilkolinesteraze (AChE), ključnog enzima za prijenos neurotransmitera acetilkolina. Uz štetan učinak na AChE, postoje dokazi o porastu oksidacijskog stresa u organizmu nakon izlaganja klorpirifosu. U ovom radu istražen je učinak niskih koncentracija klorpirifosa na biomarkere subkronične toksičnosti kod štakora. Svrha ovog i sličnih istraživanja je pridonijeti saznanjima o utjecaju pesticida na ljudsko zdravlje i okoliš. Ispitali smo štetne učinke niskih koncentracija koje su u realnim uvjetima prisutne u izloženog stanovništva, zaposlenih u proizvodnji i primjeni štetnih tvari, kao i one koje se odnose na prihvatljivi dnevni unos i referentne doze pesticida. Životinje su podijeljene u sedam skupina po pet jedinki. Četiri skupine štakora tijekom 28 dana primale su klorpirifos per os u različitim dozama:0,000374 mg/kg tjelesne težine po danu (odgovara koncentraciji eng. „residential exposure level“, REL), 0,01 mg/kg tjelesne težine po danu (odgovara koncentraciji prihvatljivog dnevnog unosa eng. „acceptable daily intake“, ADI), 0,01572 mg/kg tjelesne težine po danu (odgovara koncentraciji eng. „occupational exposure limit“, OEL) i 0,1572 mg/kg tjelesne težine po danu (odgovara koncentraciji 10xOEL)). Preostale tri skupine životinja bile su kontrolne (negativna, pozitivna i kontrola s otapalom). Istraživani parametri uključivali su mjerenje aktivnosti ukupnih kolinesteraza (ChE), acetilkolinesteraze i butirilkolinesteraze (BChE). Također, mjereni su parametri oksidacijskog stresa, razine reaktivnih kisikovih spojeva (ROS) i lipidne peroksidacije (LPO) te aktivnost antioksidacijskog enzima katalaze (CAT). Na osnovi izmjerenih vrijednosti istraživanih parametara zaključeno je da svakodnevna oralna primjena CPF-a tijekom 28 dana rezultira inhibicijom ChE, AChE i BChE, porastom razina ROS-ova i LPO te porastom aktivnosti CAT.

Organophosphate compounds are currently the most used type of pesticides. A large share of its administration involves chlopyrifos (CPF), a wide range organophosphate pesticide. It is well-known that pesticides are toxic compounds with harmful effects on human health and the environment. The main toxicity mechanism of organophospate compounds is irreversible acetylcholinesterase (AChE) inhibition, i.e. the inhibition of the primary channel for the distribution of the neurotransmitter acetylcholine. Apart from toxic effects on AChE, there is also evidence of an increase of oxidative stress in the organism, after chlorpyrifos exposure. This study investigated the effect of chlorpyrifos on biochemical markers of subchronic toxicity in rats. To investigate the influence of the pesticide on human health and the environment, we tested low concentrations that matched real exposure and were estimated on the basis of residential exposure levels, acceptable daily intake and occupational exposure limit. Animals were divided into seven groups of five animals. In four of the groups, the animals were orally administrated chlorpyrifos at different dosages (0.000374 mg/kg b.w./day (REL), 0,01 mg/kg b.w./day (ADI), 0.01572 mg/kg b.w./day (OEL) and 0.1572 mg/kg b.w./day (10xOEL)). The remaining three groups were control groups. The parameters we investigated were overall cholinesterase (ChE), acetylcholinesterase (AChE) and butyrylcholinesterase (BChE). We also measured parameters of oxidative stress, reactive oxygen species (ROS), lipid peroxidation (LPO) and catalase (CAT), the antioxidative defence mechanism. The investigated parameters showed certain harmful effects that exposure to CPF has on an organism. Oral administration of chlorpyrifos throughout 28 days resulted in ChE, AChE and BChE inhibition, increase od ROS levels and LPO, as well as with increased CAT activity.
