Abstract	Transkripcija i eksport kompetentnih mRNP-ova su povezani pomoću CTD-a (karboksi terminalne domene) RNA polimeraze II. CTD služi kao platforma za uzastopnu regrutaciju različitih proteinskih faktora. Defektni mRNP-ovi su prepoznati, zadržani i razgrađeni pomoću jezgrinog sustava kontrole kvalitete. Da bi se bolje razumjeli molekularni mehanizmi uključeni u prepoznavanje defektnih mRNP-ova, naš tim je razvio sustav kojim se može poremetiti mRNP biogeneza. NRD kompleks je jedna od glavnih komponenti jezgrinog sustava kontrole kvalitete. Asocijacija NRD-a s CTD-om omogućava prepoznavanje aberantnih transkripata i njihovu eliminaciju pomoću jezgrinog egzosoma. Ovaj način kontrole kvalitete može biti uništen ukoliko su određeni proteini uključeni u terminaciju transkripcije overeksprimirani, te spječavaju prepoznavanje/uništavanje defektnih mRNP-ova. Overeksprimirali smo nekoliko transkripcijsko terminacijskih proteina in vivo i analizirali posljedice u stanicama u kojima su defektni mRNP-ovi bili sintetizirani. Istraživanje je pokazalo da proteini uključeni u terminaciju transkripcije imaju različite uloge: overekspresija nekih od njih može poremetiti proces prepoznavanja defektivnih mRNP-ova, dok drugi ne pokazuju ovakav učinak.
Abstract (english)	Transcription and export of competent mRNPs are coupled by the CTD(Carboxy Terminal Domain) of RNA polymerase II that serves as a platform for sequential recruitment of different protein factors. Defective mRNPs are recognised, retained, and degraded by nuclear quality control. Our team have developed a system to artificially disrupt mRNP biogenesis in the cell to better understand the molecular mechanisms involved in the recognition of defective mRNPs. We have shown that one of the major components of quality control is the NRD complex whose association with the CTD allows the recognition of aberrant transcripts and their elimination by the nuclear exosome. This quality control may be destroyed if certain proteins involved in transcription termination are over-expressed, thus preventing the detection/destruction of defective mRNPs. To understand how this delicate balance between transcription termination proteins and surveillance system operates in the cell, we have over-expressed several transcription termination factors in vivo and analysed the consequences in the cells where defective mRNPs were synthesised. This research has shown that proteins involved in transcription termination have distinct roles: the overexpression of some proteins disrupts the process of recognition of defective mRNPs while the others do not show this effect.
