| Abstract | Kronična izloženost dimu cigareta oštećuje epitel dišnih putova, što dovodi do aktivacije signalnih kaskada koje potiču otpuštanje upalnih citokina, proteolitičkih enzima i reaktivnih kisikovih spojeva koji uzrokuju dodatna oštećenja. Adenozin-trifosfat (ATP) u stanici je snižen kod nekroznog umiranja stanica, dok se ATP izlučen iz stanica smatra molekularnim obrascem oštećenja i potiče upalne odgovore u stanicama. Cilj istraživanja bio je ispitati utječe li dim cigareta na koncentraciju ATP-a, odnosno preživljavanje stanica, u NCI-H292 i THP-1 staničnim linijama te potiče li lučenje ATP-a u izvanstanični prostor. Luminometrijskom metodom je izmjerena koncentracija ukupnog i izvanstaničnog ATP-a. Kako su koncentracije eATP-a jako niske, koncentracije dobivene za ukupni ATP ukazuje na promjene unutarstaničnih koncentracija ATP-a. 15 % CSE uzrokovao je smanjenu ukupnu koncentraciju ATP-a u NCI-H292 stanicama u svim vremenskim točkama za 1000 stanica, nakon 2, 4 i 24 h za 5000 stanica te nakon 24 h za 10000 stanica. 2,5 % CSE smanjio je koncentraciju ukupnog ATP-a samo nakon 24 h. 15 % CSE potaknuo je značajno lučenje ATP-a izvan stanica nakon 1 h za 5000 i 10000 stanica, te nakon 24 h za 10000 stanica, dok je 2,5 % CSE uzrokovao značajno lučenje ATP-a samo za 10000 stanica tretiranih 1 h. THP-1 stanice tretirane s 5 % CSE-a pokazale su smanjenu koncentraciju ukupnog ATP-a za 10000 stanica nakon 4 h, a tretiranje s 2,5 % i 5 % CSE nakon 24 h za 10000, 25000 i 50000, odnosno 25000 i 50000 stanica. 2,5 % CSE je kod THP-1 stanica doveo do povećanja koncentracije ATP-a kod 50000 stanica nakon 4 h, te 5 % CSE nakon 2 h za 10000 stanica. Povećanje koncentracije izvanstaničnog ATP-a 5 % CSE potaknuo je za 10000 stanica nakon 4 h, a 2,5 % i 5 % CSE kod 25000 nakon 1 h. Kod obje vrste stanica tretiranje tijekom dužeg vremena dovodi do smanjenja koncentracije izvanstaničnog ATP-a, što je vjerojatno uzrokovano aktiviranjem ATP-aza. Učinak CSE-a na koncentraciju unutarstaničnog i izvanstaničnog ATP-a ovisi o njegovoj koncentraciji, vremenu izloženosti stanica CSE-u, te o vrsti i broju stanica. Povećanje unutarstaničnog ATP-a ukazuje na proliferacijski učinak CSE-a, smanjenje unutarstaničnog ATP-a ukazuje na citotoksičnost CSE-a, a povećanje izvanstaničnog ATP-a ukazuje na upalno djelovanje CSE-a. |
| Abstract (english) | Chronic exposure to cigarette smoke damages airways epithelia leading to activation of signaling cascades that promote the release of pro-inflammatory cytokines, proteolytic enzymes and reactive oxygen species that cause additional damage. Cellular adenosine triphosphate (ATP) is reduced during necrosis, while the ATP secreted from cells is considered as damage-associated molecular pattern who promotes inflammatory responses of cells. The aim of this study was to explore if cigarette smoke has an effect on ATP intracellular concentration as well as if it promotes ATP release from NCI-H292 or THP-1 cells to extracellular milieu. Luminometric method was used to measure concentration of total and extracellular ATP. Because eATP concentration is very low, concentration obtained for total ATP mirror the changes of intracellular ATP concentration. 15% CSE caused a reduced total ATP concentration in NCI-H292 cells in all time points for 1000 cells, after 2, 4 and 24 h for 5 000 cells and after 24 h for 10 000 cells. 2.5% CSE decreased total ATP concentrations only after 24 h. 15% CSE induced significant ATP release from cells after 1 h for 5 000 and 10 000 cells, and after 24 h for 10 000 cells, while 2.5% CSE caused significant ATP secretion only for 10 000 cells when applied for 1 h. THP-1 cells treated with 5% CSE showed a decreased concentration of total ATP for 10 000 cells after 4 h and treatment with 2.5% and 5% CSE after 24 h for 10 000, 25 000 and 50 000, 25 000 and 50 000 respectively. 2.5% CSE in THP-1 cells resulted in an increase in ATP concentration for 50 000 cells after 4 h and 5% CSE after 2 h with 10 000 cells. 5% CSE stimulated increase in extracellular ATP concentration at 10 000 cells after 4 h, while 2.5% and 5% CSE caused significant increase of eATP at 25 000 cells after 1 h.
In both cell lines longer treatment leads to a reduction of the extracellular ATP concentration, which is probably caused by the activation of ATPases. The effect of CSE on the concentration of intracellular and extracellular ATP depends on its concentration, duration of cell exposure to CSE, cell type and cell number. Increase in intracellular ATP indicates the proliferative effect of CSE, decrease in intracellular ATP indicates cytotoxicity of CSE, while increase in extracellular ATP indicates the pro-inflammatory effect of CSE. |
