Abstract	Fibromialgija je česti, idiopatski, i kompleksni kronični sindrom koji je karakteriziran alodinijom. Češće obolijevaju žene u dobi 20-50 godina, ali se bolest javlja i kod muškaraca te žena ostalih dobnih skupina. Etiologija fibromialgije je nejasna. Brojna, istraživanja ukazuju na razne faktore koji utječu na nastanak samoga sindroma, poput: disfunkcije središnjega i autonomnoga živčanoga i imunosnoga sustava te genski i psihosocijalni utjecaji. Primarni simptom fibromialgije jest kronična bol na više mjesta na tijelu. Osim toga simptoma često su prisutni još umor, poremećaj spavanja, glavobolje, parestezija i dr. Trenutno specifični pokazatelji fibromialgije. Iz tog razloga, fibromialgija se dijagnosticira pomoću ACR-ovih kriterija. E-selektin je protein koji pripada obitelji selektina. Selektini vežu ugljikohidrate. E-selektin je glikoprotein koji se aktivira pod utjecajem citokina, IL-1, TNF-a i dr. E-selektin je uključen u aktivaciju stanica imunosnog sustava, T-limfocita i trombocita. Nedostatak selektina može imati ozbiljne posljedice kod ljudi, poput primjerice sklonosti određenim bolestima te povećanoj vjerojatnosti recidiva bakterijskih infekcija. Razina E-selektina povećana je kod određenih patoloških stanja, posebice kod tumora i nekih upalnih bolesti. Neka istraživanja ukazuju da su koncentracije E-selektina kod osoba koje boluju od fibromialgije povišene. Glavni cilj ovoga diplomskog rada bio je utvrditi značajnost koncetracije E-selektina za dijagnostiku fibromialgije. Metoda koja je korištena u ovom diplomskom radu je imunokemijska ELISA-metoda. Dobiveni rezultati ukazuju da je koncentracija E-selektina u skupini oboljelih od fibromialgije niže u odnosu na skupinu zdravih dobrovoljaca. S obzirom da to sniženje nije statistički značajno može se zaključiti da se E-selektin ne bi mogao koristiti kao specifični biomarker za fibromialgiju.
Abstract (english)	Fibromyalgia is an idiopathic, common and complex chronic syndrome characterized by allodynia. It occurs more frequently in women aged 20-50 years, but also occurs in men. The etiology of fibromyalgia is unclear, but some studies have been indicated various factors that influence the occurrence of the syndrome, such as: dysfunctions of the central and autonomic nervous system, dysfunctions of the immune system and the genetic and psychosocial influences. The primary symptom of fibromyalgia is a chronic pain in several places at the body. Besides that, other symptoms include fatigue, sleep disturbances, headaches, paresthesia and others. Recently, fibromyalgia has been diagnosed by ACR criteria, because there is no laboratory or objective tests for its diagnosis. E-selectin is a protein that belongs to selectin family of proteins. Selectins bind carbohydrates. E-selectin is a glycoprotein activated by cytokines, IL-1, TNF-a, etc. E-selectin is involved in activation of immune cells, T-lymphocytes, and platelets. Lack of selectins could have serious consequences, such as the tendency to certain diseases, and increased chance for recurrence of bacterial infections. The level of E-selectin is increased in certain pathological conditions, particularly in some types of carcinoma and some inflammatory diseases. There are studies that suggest the possibility of increased concentrations of E-selectin in people who suffer from fibromyalgia. The main objective of this master thesis was to determine the significance of the concentration of E-selectin for the diagnosis of fibromyalgia. The method that was used in this thesis was ELISA method. The results indicate that the concentration of E-selectin is not relevant for the diagnosis of fibromyalgia.
