Abstract
Cilj rada bio je odrediti kako prehrana bogata kolesterolom utječe na koncentraciju kolesterola i razinu oksidativnog stresa u jetri i mozgu miševa (C57BL). Također, cilj rada je bio na staničnom modelu CHOwt i NPC1null (stanice CHO s deletiranim genom za protein NPC1 koje nakupljaju kolesterol) odrediti antioksidativnu obranu stanica pri normalnim uvjetima i u uvjetima povećanog oksidativnog stresa. U pokusu je mjerena lipidna peroksidacija, koncentracija ukupnog glutationa te aktivnost antioksidativnih enzima superoksid dismutaze i katalaze. Rezultati su pokazali da je mozak zaštićeniji od jetre od oksidativnog stresa nastalog kao posljedica hiperkolesterolemije. Stanice NPC1null imaju povećani oksidativni stres u normalnim uvjetima. Pri povećanom oksidativnom stresu stanice NPC1null imaju oslabljenu antioksidativnu obranu. Na životinjskom i staničnom modelu je pokazano da su na oksidativni stres uzrokovan nakupljanjem kolesterola najosjetljiviji mitohondriji, a najpogođeniji antioksidans je superoksid dismutaza.
Abstract (english)
Aim of this study was to determine how cholesterol-rich diet influences the cholesterol concentration and oxidative stress levels in mice (C57BL) liver and brain. Also, aim was to determine the levels of antioxidant defense systems in cell model CHOwt and NPC1null (CHO cells with deleted gene for NPC1 protein which accumulate cholesterol) in normal conditions and under higher levels of oxidative stress. In this study the amount of lipid peroxidation, concentration of total glutathione and activity of antioxidant enzymes superoxide dismutase and catalase was measured. Results have shown that the brain is more protected from oxidative stress caused by hypercholesterolemia than the liver. Cells NPC1null have higher oxidative stress levels in normal conditions. Under conditions of higher oxidative stress, cells NPC1null have lower antioxidant defense. In animal and cell models, mitochondria are especially susceptible to oxidative stress caused by cholesterol accumulation and the most affected antioxidant is superoxide dismutase.
