Abstract	Newcastleska bolest (NB) je izuzetno kontagiozna virusna zarazna bolest kokoši i drugih vrsta ptica. Kontrola bolesti se provodi cijepljenjem i nespecifičnim mjerama. Kako bi se izmjerila učinkovitost postojećeg cjepnog programa na proizvodnoj farmi može se paralelno koristiti procjena humoralne i stanične imunosti. Cilj ovog rada je opisati metode kojima se može odrediti stanična imunost u određenim vremenskim razmacima od cijepljenja i time njeno trajanje. Stanična imunost određuje se mjerenjem in vitro proliferacije imunosnih stanica nakon stimulacije uz određivanje razine citokinskog odgovora.Od metoda mogu se koristiti protočna citometrija, imunoenzimni test, ELISPOT test, a od molekularnih metoda lančana reakcija polimerazom u stvarnom vremenu. U rutinskoj dijagnostici kao najprikladnija metoda pokazao se PCR zbogjednostavnosti i brzine izvedbe. Stanični imunosni odgovor je veoma bitan za ranu i cijepljenjem potaknutu zaštitu organizma, međutim sam nije dostatan da zaštiti ptice od VNB.Dostupnaistraživanja ne ukazuju na stvarno stanje in vivo te je potrebno provesti dodatna istraživanja.
Abstract (english)	Newcastle disease (ND) is very contagious viral disease of poultry and other birds. Control of ND is achieved by vaccination and non-specific measures. The efficiency of vaccination programs implemented on farms is assessed by testing humoral and cell-mediated immunity (CMI). The aim of this paper was to describe the methods used for determination of CMI in certain time points after vaccination (dpv), and thus, duration of CMI. Cell-mediated immunity can be determined by in vitro stimulation and proliferation of peripheral blood mononuclear cells (PBMCs), and the level of cytokine response/release. Flow cytometry, ELISA, ELISPOT, and Real -Time PCR are mostly used in CMI assessment. Real-Time PCR has been proven to be the most appropriate method for routine diagnostic, due to simplicity and speed of the procedure. CMI plays a crucial role in early immune response following immunisation. However, it's not sufficient to protect by itself the birds from NDV infection. Available studies so far don't reveal the true in vivo status, therefore further research is required.
