Abstract
Fitopatogena gljiva Cryphonectria parasitica uzročnik je raka kore pitomog kestena Castanea sativa. Zaraza gljive hipovirusom, Cryphonectria hipovirus 1 (CHV1), smanjuje virulentnost i reproduktivni kapacitet gljive što doprinosi biološkoj kontroli raka kestenove kore. Hipovirulentna gljiva zaražena CHV1 virusom, unosom u aktivni rak uzrokovan virulentnim sojem gljive C. parasitica, omogućuje formiranje kalusa i zaustavlja širenje raka. Cilj ovog istraživanja bio je karakterizirati uzorke gljive C. parasitica na osnovi morfologije, tipa vegetativne kompatibilnosti (VC) i molekularnih analiza. Rezultati morfologije pokazali su prisutnost 55% žutih virulentnih sojeva i 45% bijelih sojeva gljive zaraženih hipovirusom. Najzastupljeniji vc tip gljive bio je EU2 (37%) zatim EU12 (27%), EU5 (18%) i EU1 (9%) dok je 9% bilo neodređenog VC tipa. U hipovirulentnim sojevima gljive dokazana je prisutnost hipovirusa CHV1 izolacijom dvolančane hipovirusne RNA i RT-PCR metodom, dok je analiza polimorfizma duljine restrikcijskih odsječaka (RFLP) potvrdila da CHV1 pripada talijanskom podtipu, CHV1-I.

Phytopathogenic fungus Cryphonectria parasitica is the causal agent of chestnut blight. Cryphonectria hypovirus 1 (CHV1) infects the fungal pathogen and reduces virulence and reproductive capacity of fungus which is important in the biological control of chestnut blight. If a hypovirulent fungal strain carrying CHV1 is introduced into an active canker caused by virulent C. parasitica strain, canker expansion ceases and callus is formed. The aim of this study was to characterize isolates of the fungus C. parasitica on the basis of morphology, type of vegetative compatibility and molecular analysis. Morphology results showed 55% yellow and 45% white hypovirulent strains. The most common VC type is the EU2 (37%) followed by the EU12 (27%), EU5 (18%) and EU1 (9%), while 9% were indeterminate VC type. Hypovirus CHV1 was found in all tested hypovirulent strains by dsRNA isolation and RT-PCR method, and the analysis of restriction fragment length polymorphism (RFLP) confirmed that CHV1 isolates belong to Italian subtype, CHV1-I.
