Abstract
Urin predstavlja značajnu tjelesnu tekućinu pri dijagnosticiranju oboljenja. U većini slučajeva bakterije, koje koloniziraju urinarni trakt, ne uzrokuju nastanak bolesti već se imunološki sustav organizma na njih prilagodi. Međutim, neke bakterije uzrokuju nastanak bolesti i zadržavanje unutar urinarnog trakta. Infekcije urinarnog trakta jedne su od najčešćih infekcija zemalja u razvoju od kojih godišnje oboli više od 250 milijuna ljudi. S ciljem identifikacije mikroorganizama, u ovom radu izolirani su proteini bakterija prisutni u urinu koji su potom pročišćeni, fragmentirani i frakcionirani. Za identifikaciju proteina i vrste mikroorganizma korištena je metoda temeljena na proteomskom pristupu koja uključuje primjenu spektrometra masa MALDI-TOF/TOF i bioinformatičkog alata ProteinReader. Cilj ovog rada je bio usporediti rezultate dobivene mikrobiološkom analizom uzoraka urina s onima dobivenim analizom na spektrometru masa. Rezultati proteomskog istraživanja odgovaraju rezultatima mikrobioloških analiza, iako su prvi specifičniji te daju točan soj mikroorganizma, a sama analiza je puno brža.
Abstract (english)
Urin represents a significant body fluid in diagnosing the disease. In most cases bacteria which colonize the urinary tract do not cause the disease because the host immune system adjusts to it. However, some bacteria cause development of the disease and the retention within the urinary tract. Urinary tract infections are one of the most common infections in developing countries, with an estimated annual global incidence of at least 250 million. With the aim of identifying microorganisms, proteins from urine bacteria were isolated, purified, fragmented and fractionated. A proteomic approach was used to identify proteins and microorganisms, which included the application of the MALDI-TOF/TOF mass spectrometer and the bioinformatic tool ProteinReader. The aim of this study was to compare the results obtained by microbiological analysis of urine samples with those obtained by analysis on the mass spectrometer. The results of the proteomic research correspond to the results of microbiological analyzes, although the first ones are more specific and provide the exact microorganism strain, and the analysis itself is much faster.
