Abstract
Arsen spada u skupinu teških metala i u okolišu se javlja u nekoliko oksidacijskih stanja, pri čemu su arsenat i arsenit najčešći oblici. Toksičnost, mobilnost i raspoloživost arsena u okolišu ovise o njegovom oksidacijskom stanju. Arsen se otpušta u okoliš kao rezultat geogenih i antropogenih aktivnosti. Toksičnost arsena postao je jedan od globalnih problema diljem svijeta, s obzirom da se otpušta u vode i tla gdje dolazi do kontaminacije. Izlaganje arsenu može izazvati promjene na razini stanice kao što su inaktivacija enzima uključenih u DNA replikaciju i popravak, sintezu nukleinskih kiselina i fosfolipida, te na razini organizma npr. rak kože, mučnina, povraćanje, svrbež itd. Iako je toksičan za većinu organizama, neki prokarioti su razvili mehanizme za dobivanje energije putem redukcije ili oksidacije arsena. Upravo su mikroorganizmi uključeni u kruženje arsena u prirodi, utječući na njegov biokemijski ciklus, na način da pretvaraju arsen u oblike različite topljivosti, mobilnosti, raspoloživosti i toksičnosti. Glavni mehanizmi kojim bakterije mijenjaju ove značajke su reakcije oksidacije, redukcije i metilacije. Zahvaljujući ovim sposobnostima, bakterije se mogu koristiti za smanjivanje koncentracije arsena u okolišu. Razumijevanje raznolikosti i rasprostranjenosti bakterija rezistentnih na arsen te mehanizama koji im omogućuju rezistenciju može doprinijeti uspješnoj bioremedijaciji na područjima kontaminiranim arsenom.
Abstract (english)
Arsenic is a heavy metal occurring in several oxidation states with arsenate and arsenite being the most common forms. Arsenic toxicity, bioavailability and mobility depend on its oxidation state. It is released into the environment as the result of geogenic and anthropogenic activities. Arsenic toxicity has become a global concern due to increasing contamination of water and soil in many regions of the world. Exposure to arsenic causes changes both on the cellular level, i.e. inactivation of enzymes involved in DNA replication, repair, nucleic acid and phospholipid synthesis, and on the level of the whole organism, i.e. skin cancer, nausea, vomiting, itching etc. Although As is toxic to most organisms, some prokaryotes have developed mechanisms to produce energy by oxidizing or reducing it. Therefore, microorganisms are involved in the arsenic cycle by influencing its biochemical cycle through conversion to forms with different solubility, mobility, bioavailability, and toxicity. These bacteria may be used for removal of arsenic from contaminated environment. Understanding the diversity and distribution of arsenic-resistant bacteria and the mechanisms involved in arsenic-resistance could contribute to successful bioremediation of arsenic-contaminated areas.
