Abstract
Zadatak rada bio je utvrditi povezanost porođajne mase i gestacijskih parametara s izloženošdu ohratoksinu A (OTA) putem hrane. Analizirani su uzorci prvog jutarnjeg urina 40 ispitanica, od kojih je 20 bilo iz ruralne, a 20 iz urbane sredine. OTA i njegov metabolit ohratoksin α (OTα) određeni su HPLC metodom nakon tekućinsko-tekućinske ekstrakcije. Izvršena je i enzimska (β-glukuronidaza i arilsulfataza) hidroliza konjugata. Ispitanice su popunile kratak upitnik o učestalosti konzumacije određenih namirnica (FFQ) kako bi se procijenio unos OTA hranom koji je koreliran s količinom OTA i OTα u urinu. Za razliku od OTA, srednja razina OTα se višestruko povedala nakon enzimskog tretmana. Utvrđen je trend negativne povezanosti, koji nije bio statistički značajan, mase i duljine novorođenčeta i urinarnih koncentracija OTA i OTα. Utvrđen je i nešto vedi dnevni unos OTA ispitanica srednjoškolskog obrazovanja, kao i vede koncentracije OTα u urinu ispitanica srednjoškolskog obrazovanja, te onih najboljeg materijalnog statusa. Sukladno radovima drugih autora, rezultati upućuju na vezu izloženosti i unosa specifičnih namirnica, iako bi detaljno poznavanje izvora namirnica moglo biti presudno u karakterizaciji rizika za fetus.
Abstract (english)
The aim of this work was to determine relationship between birth weight and gestational parameters with dietary exposure to ochratoxin A (OTA). First-void urine samples of 40 subjects, of which 20 was from rural, and 20 from urban area, were analyzed. OTA and its major metabolite ochratoxin α (OTα) were determined by HPLC-FLD following liquid-liquid extraction. Enzymatic hydrolysis (β-glucuronidase/aryl sulfatase) of conjugates was also performed. The subjects filled a short food frequency questionnaire (FFQ) to assess the OTA intake and to correlate it with OTA and OTα urinary concentrations. Unlike OTA, OTα levels increased significantly following enzymatic treatment. A trend of negative correlation was determined, although not statistically significant, between newborn weight and length and urinary OTA and OTα concentrations. Slightly higher daily OTA intake and urinary OTα concentrations were determined in high school educated subjects. Similarly, nonsignificantly higher OTα urinary concentrations were estimated in well-off women. In agreement with other studies, the results indicate a relationship between exposure to OTA and intake of specific foods. Nevertheless, a more detailed knowledge of food sources may be critical in fetal risk characterization.
