Abstract
Čaj je jedan od najpopularnijih napitaka zbog njegovog ugodnog okusa i pozitivnih učinaka na zdravlje. Produkti biljnih čajeva izazivaju sve veću pažnju zbog njihovih pozitivnih zdravstvenih učinaka te velike koristi u farmaceutskoj i poljoprivrednoj industriji. Konzumacija čaja, osobito zelenog čaja, povezana je sa niskom učestalošću kronično patoloških bolesti u koje je uključen oksidativni stres, poput raka i kardiovaskularnih bolesti. Zeleni čaj može zaštititi organizam od razvoja malignih, kardiovaskularnih i metaboličkih bolesti. Farmakološki najzanimljiviji spojevi zelenog čaja su polifenoli. Polifenoli posjeduju antioksidativna, protuupalna, hipoglikemijska, hipolipidemijska svojstva, te mogu smanjiti hipertenziju. Polifenoli također imaju i antivirusna svojstva, te se vjeruje da pomažu u zaštiti od virusa gripe. Sve u svemu, zeleni čaj posjeduje mnoga bitna svojstva za očuvanje zdravlja organizma, te posljedično tome postaje sve zanimljivija meta istraživanja. Cilj ovog diplomskog rada bio je testirati utjecaj 18 spojeva zelenog čaja i 3 biljna ekstrakta na oksidativni stres i na biološki važne enzime: acetilkolinesterazu (AChE), butirilkolinesterazu (BChE) te glutation-S-transferazu (GST). Također, testirali smo i utjecaj inkapsulacije biološki važnih spojeva u liposome na inhibiciju AChE i GST. Rezultati su pokazali da 1 od testiranih spojeva posjeduje antiacetilkolinesterazno i antibutirilkolinesterazno djelovanje, dok 4 spoja zelenog čaja i 3 biljna ekstrakta posjeduju antiglutation-S transferazno djelovanje. Rezultati testiranja inkapsuliranog kvercetina u liposome pokazali su da je kvercetin zadržao svoje antiacetilkolinesterazno djelovanje, dok (-)-epigalokatehin galat inkapsuliran u liposome nije zadržao svoje antiglutation-S transferazno djelovanje. Rezultat testa antioksidativnosti pokazao je da 18 od 20 testiranih spojeva posjeduje antioksidativna svojstva.

Tea is one of the most popular bevereges because of its pleasant taste and positive effects on human health. Natural products from herbal tea have caused great interest since reasearches discovered their benificial health effects, and their use in pharmaceutical and agricultural industries. The consumption of tea, especially green tea, is associated with low incidence of chronic pathological diseases where oxidative stress is involved, for exemple cancer or cardiovascular diseases. Green tea may protect organisam from cancer, cardiovascular and metabolic diseases. Polyphelons are pharmacologically the most interesting compounds of green tea. Polyphenols have antioxidat, anti-inflammatory, hypoglycemic, and hypolipidemic properties, and may reduce hypertension. Polyphenols also have antiviral properties, which are believed to protect against influenza virus. Overall, green tea has many properties which are important for perserving health of the organism, and consequently are a very interesting reasearch target. The aim of this thesis was to test the effect of 18 compounds isolated from green tea, and 3 herbal extracts, on oxidative stress and on biologically important enzymes: acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and glutathione-S-transferase (GST). Also, we tested the effect of encapsulation of biologically important compounds in liposomes on their ability to inhibit AChE and GST. The results showed that one out of 21 tested compounds inhibits AChE and BChE, while 4 compounds from green tea and all of tested herbal extracts inhibit GST. Encapsulation of quercetin in liposomes showed that quercetin kept his ability to inhibit AChE, while (-)-epigallocatechin gallate encapsulated in liposomes loose its ability to inhibit GST. Results on the antioxidant ability showed that 18 out of 20 tested compounds possess antioxidant properties.
